Cox-2 Inhibitors

aFarmakoloji AD, Ege Üniversitesi Tıp Fakültesi, İZMİR The primary action of nonsteroidal antiinflammatory drugs (NSAIDs) is to inhibit cyclooxygenase (COX) enzyme, responsible for the production of prostaglandins. COX-1, the constitutive isoform, is involved in cell-cell signalling and tissue homeostasis. COX-2 is induced in inflammatory cells and responsible for the production of the prostanoid mediators of inflammation. The unwanted effects, especially gastrointestinal side effects, of NSAIDs which nonselectively inhibits both cyclooxygenase isoforms, are mostly due to inhibition of COX-1. The NSAIDs which selectively inhibit COX-2 have been developed in order to decrease the unwanted effects. Neither rofecoxib or celecoxib significantly interferes with platelet function, platelet aggregation or prolongs bleeding time.
